Transitional B cells represent a crucial stage in the B cell developmental pathway as these cells pass through additional check points for self tolerance during migration from bone marrow to the periphery. Akin to mouse data, our lab recently delineated multiple subsets within the human transitional B cell compartment. My research theme aims at understanding the molecular nature of these transitional subsets and investigates abnormalities, if any, in autoimmune subjects. Techniques include repertoire analysis by spectratyping, V(D)J gene usage and mutational analysis utilizing the single cell RTPCR approach and gene expression profiling by micro array. Furthermore, the immunologic effects of targeted biologics on transitional B cells are also studied in the treatment of autoimmune disorders.
