Submitting Your Samples
The URGRC offers 4 options for DNA sequencing sample submission:
Standard (Preferred)
- You supply the DNA template for sequencing and we perform the reaction using our standard primers.
If you supply custom primers for sequencing, please submit them using the "Pre-mixed" option below.
- DNA should be diluted with sterile RNase/DNase-free water.
- 0.6ml or 1.5ml eppendorf tubes are preferred but we will accept strip cap PCR tubes if properly labeled. Please do not submit multiple 0.2ml PCR tubes loosly in a ziploc bag.

Size |
Quantity |
Volume |
| Plasmid DNA | <10kb | 400ng | 10ul |
| Plasmid DNA | >10kb | 2000ng | 20ul |
| PCR Product | 100-200bp | 20ng | 10ul |
| PCR Product | 200-500bp | 40ng | 10ul |
| PCR Product | 500-1000bp | 80ng | 10ul |
| PCR Product | 1000-2000bp | 100ng | 10ul |
| PCR Product | >2kb | 400ng | 10ul |
**We ask that you please send in the volumes stated above in order to have enough material to repeat reactions if necessary.**
Pre-mixed (Preferred)
- You supply the DNA template and primer already mixed together.
- DNA/primer mix should be diluted with sterile RNase/DNase-free water.
- 0.6ml or 1.5ml eppendorf tubes are preferred but we will accept strip cap PCR tubes if properly labeled. Please do not submit multiple 0.2ml PCR tubes loosly in a ziploc bag.

Seq Drop Off
- A.) You perform the BigDye labeling reaction in your lab and send the sample to us for post-reaction cleanup. BigDye can be ordered through the URGRC by following this link.
- B.) You can also perform the post reaction cleanup in your lab using Princeton Separations Centri-Sep spin columns or Abgene Dye Terminator Removal Plates. The samples should then be dried down in 1.5ml eppendorf tube and sent to us for sequencing.
**Please be sure to select YES for option A and NO for option B in the"Sequencing Reaction Clean-up" field when you enter the sample information on dnaLIMS.**
NanoDrop Spectrophotometer
The NanoDrop is a free service offered by URGRC and is located in the Medical Center, room G-7415. Please see the user manual for more detailed information.
