Signal Transduction by GPR56

Two Activation States of GPR56.

Through the course of analyzing GPR56 function in melanomas, we established a cellular system to investigate its signaling pathway. Our results proposed that the receptor exists in different activation states. The full-length N-terminus fragment (NTF) acts as an inverse agonist to inhibit the basal activity of the seven transmembrane domains (CTF), but the truncated N-terminus (ΔSTP-NTF) acts as an agonist to activate CTF (see above figure). We will extend the above study to delineate the signaling pathway mediated by GPR56 and TG2. Our first line of investigation will be to identify the components that transmit the activation states of GPR56 and their responses to ligand stimulation.
