RTOG 0424: A Phase II Study of a Temozolomide-Based Chemoradiotherapy Regimen for High Risk Low-Grade Gliomas
- Brief Overview:
Eligible patients will undergo radiation therapy along with oral temozolomide chemotherapy followed by chemotherapy alone.
- Inclusion Criteria
- Histological proof of a unifocal or multifocal supratentorial WHO grade II astrocytoma (diffuse fibrillary, protoplasmic, or gemistocytic), oligodendroglioma, or oligoastrocytoma
- Patient must have at least three of the following risk factors:
- Age greater than or equal to 40 years
- Largest preoperative diameter of tumor greater than 6 cm
- Tumor crosses midline of brain
- Tumor subtype of astrocytoma (astrocytoma dominant)
- Preoperative Neurological function status greater than 1
- Exclusion Criteria
- Prior chemotherapy
- Prior radiation to the brain or head/neck
- Known HIV infection
- Active infectious process
