My research is focused on the regulation of mRNA stability of monocyte chemoattractant protein-1 (MCP-1) by PDGF and Angiotensin II (Ang II) in smooth muscle cells (SMC). MCP-1 promotes atherosclerosis and is a mediator of the response to arterial injury. MCP-1 level tracks its mRNA level under physiological or pathological conditions in animal models. MCP-1 mRNA is markedly elevated in response to PDGF-B or Ang II elevation largely due to increase of mRNA stability. Both PDGF-B and Ang II stabilize MCP-1 mRNA through a PKCd-dependent signaling pathway. Our ongoing research is to fully figure out the signaling pathways mediating the mRNA stabilizing effect, to isolate the MCP-1 mRNA stabilizing factor induced by PDGF-B and Ang II, and to reveal the RNases involved in PDGF-b and Ang II induced MCP-1 mRNA stabilization.
