Functional genomics for HIV host factors.

To produce a comprehensive map of host-virus interactions, we applied a screening strategy to complete parallel siRNA screens using multiple orthologous RNAi reagents (MORR). We also successfully developed an analytic pipeline by integrating multiple established bioinformatic and biostatistic algorithms, including RIGER and GESS, to comprehensively evaluate raw screening results and filter false-positive hits. We find this approach is useful to overcome the current hurdles confronting functional genomics. We are currently initiating a focused siRNA screen for a set of host factors that particularly target viral transcription and latency (HIV, EBV, and KSHV).
