Flow Analysis

RHIC flow cytometry analysis instrumentation is integrated into the URSMD Flow Cytometry Core Facility providing quality control, maintenance, and calibration services for flow users. Instruments available in the Flow Core can be viewed using the analysis page. To view user guidelines and billing & fee structure, please view the Billing & Fee Structure page. The RHIC 11-color and Flow Core 12-color are configured similarly except for an additional PMT on the violet laser that may be used with Q-dot 605. Any of the instruments may be reserved by registered flow users by following the online calendar link on the left. To become a registered user, fill out the new user form located on the Flow Core website. You will be contacted to schedule time for training before being given a username and password to access specific analysis instruments and the online reservation system. If you have additional questions, please contact Tim Bushnell (x3-5535) or Matt Cochran (x3-4473).

The following instruments are physically located in the RHIC Core Lab (MRB 2-9905):

Billing information and fee structure for the 18-color and 11-color LSRII can also be found on the Flow Core website's Billing & Fee Structure page.

When all considerations are equal, the RHIC encourages users to purchase BD reagents for flow cytometry since this supports URMC/BD instrument rental contracts.

Flow Sorting

The RHIC designs, develops, and validates cell sorting strategies using a 7-color FACSAria located in the CVBI or a 13-color FACSAria located in the Flow Core facility. Please contact the RHIC lab director, Sally Quataert (3-2454), if you are interested in collaborative cell sorting projects or for use of existing panels.

For rules and guidelines pertaining to the FACSAria located in the CVBI, click on the CVBI Cell Sorting Facility website. For the FACSAria located in the Flow Core facility, please click on the URMC Flow Core website.

Luminex

Luminex Xmap Multiplexed microbead array technology: Researchers interested in taking advantage of new technologies to analyze smaller sample volumes for multiple analytes simultaneously have access to the technology in the RHIC core lab. The Luminex microbead array system is applicable for a variety of small molecule or biomolecules including nucleic acid, protein and carbohydrate. The RHIC lab sponsors demonstrations and assists investigators in using Luminex microarray bead technology. The RHIC maintains a Luminex 200 core service with a BioRad BioPlex instrument. The BioRad BioPlex offers both user-friendly software and calibration/validation and quality control options suitable for clinical study and research use. The Bioplex 200 is located in room 3-4118.

To use the Luminex service, you need to register and be trained. There is a $100 training fee. Please complete the Luminex Training Request Form to register and contact Sally Quataert to be trained. Once this is done, you will have access to the instrument calendar to sign up for use. There is an $18/hour fee for Luminex use.

For further information, please contact Sally Quataert at sally_quataert@urmc.rochester.edu or by phone at x3-2454.

ELISPOT Reader

CTL ReaderThe CTL Immunospot® S2A Core Analyzer is an automated ELISPOT reader for ELISPOT work in 96 well format. ELISPOT data may be transferred to the RHIC server for secure backup and access. A satellite computer is available with C.T.L. Immunospot® for data analysis.

The ELISPOT Reader is located in MRB 3-9878. A hard copy sign-up log is located next to the instrument. Reader access accounts are granted after being trained by Dr. Ernest Wang.

Please contact Dr. Wang at jyhchiang_wang@urmc.rochester.edu.
