Research Overview

Clinical studies on a novel inhibitor of influenza virus (GG167): GG 167 is an inhibitor of the influenza virus neuraminidase, which binds to the active site of the enzyme with high affinity. GG167 was designed partly based on the crystal structure of the influenza virus neuraminidase and is therefore often cited as an example of "rational" drug design. Because the drug is designed to interact with a highly conserved region of the neuraminidase, it has been theorized that drug resistant mutants would be isolated infrequently. So far, this seems to be true. Viruses which are resistant to the inhibitory effects of GG167 can be isolated relatively easily in cell culture. However, GG167 remains active in animals infected with these "resistant" mutants. This is because the mechanism of in vitro drug resistance appears to involve second site mutations in the hemagglutinin gene which allow the virus to utilize a method of egress from the cell that is not available in vivo. Truly resistant viruses have not been seen in clinical studies so far. To date, we have done three studies of GG167 in humans. These have included human challenge studies with infectious influenza A virus; our studies remain ongoing.

Other Studies: Other studies are centered on clinical studies to evaluate experimental measures to control viral diseases, particularly influenza. The laboratory component of this is applied laboratory studies to support the clinical vaccine and drug trials.
