Xenogen IVIS In Vivo Imaging Core Director Randall Rossi, M.S. Mission Statement The mission of the in vivo bioluminescence imaging core is to allow the detection of bioluminescent tracer molecules in living small animals. Statement of Core Services/Functions The core provides training in the use of a Xenogen IVIS bioluminescent imager, and an introduction to the software used to analyse data generated by this instrument. We then charge for access to the instrument, based on the time used. All users are charged the same rate. Users must arrange for mice that contain bioluminescent molecules to be located in the B.9856 animal suite in the KMRB vivarium. This is a one way, SPF suite that allows the use of low-grade mouse pathogens and vectors that are not readily spread from animal to animal. Mice are anesthetized, and given an injection of the bioluminescent substrate (usually Luciferin). The mice are placed in a light-tight chamber, and imaged using a sensitive CCD camera. The proprietary Living Image software allows the emitted light to be superimposed on a digital image of the mice, regions of interest to be defined, and photon flux to be measured. Specific Instrumentation/Equipment The Xenogen IVIS Imager from Caliper Lifesciences is a first-generation instrument that documents bioluminescence across the visible spectrum. Multiparameter imaging is possible in principle, but would require the use of bioluminescent reporters that depend on different substrates. In practical terms, it is a 1-parameter instrument.
