HBMI Core Equipment and Facilities

The HBMI Core is located in rooms 1.7574, 1.7564, 1.7534, and the surgery rooms - 1.7529 and 1.7538. of the Center for Musculoskeletal Research on the University of Rochester Medical Center campus. The Core occupies nearly 1500 square feet of laboratory and office space that houses integral equipment for each HBMI Sub-Core.

Histology, IHC and ISH Sub-Core

IHC ISH Sub-CoreThe Histology, IHC, and ISH Sub-Core The Histology, IHC, and ISH Sub-Core provides resources and support for the generation of tissue sections from paraffin and frozen embedded tissue samples and the subsequent histological, IHC, and ISH analyses. Major equipment includes:

  • Sakura Tissue-Tek VIP 6 automated histoprocessor (1)
  • Sakura Tissue-Tek TEC 5 paraffin embedding center (2)
  • Microm HM355S automated microtome (3)
  • Leica RM2255 automated microtome (4)
  • Two Microm HM315 microtomes & one Microm HM325 microtome (5)
  • Leica CM1850 cryostat with a CryoJane tape transfer system (6)
  • Leica CM1860UV cyrostat (7)

Microscopy, Histomorphometry, and Imaging Sub-Core

The Microscopy, Histomorphometry, and I Imaging Sub-Core provides resources and support for the generation of digital bright field, dark field, and fluorescent imaging of tissue or cell preparations and the subsequent quantitative and histomorphometric image analyses.

Olympus VisiopharmMajor equipment includes:

  • Olympus VS120 Virtual Slide Microscope and Visiopharm Image Analysis System (1)
    The system features include:
    • 100 slide capacity with a dependable and durable slide loader
    • Diagnostic quality images produced using 2x, 10x, 20x or 40x objective lenses
    • 2/3” CCD camera (3.45 μm x 3.45 μm pixel size, high sensitivity, high resolution)
    • Macro image for complete overview of the glass slide
    • Entire slide scan or a region of interest scan
    • Fluorescence filters included are FITC; Texas Red; Triple with FITC, Texas Red, & DAPI, with reduced bleaching problems in fluorescence slides
    • Z-plane focus available on Brightfield and Fluorescence slides
  • Microscopy, Histomorphometry and Imaging Sub-CoreTwo Zeiss Axioskop 40 BF/DF/fluorescent microscopes with SPOT RT3 color/slider cameras and dedicated computer and software packages (2)
  • Olympus SZX12 Stereoscope with a SPOT Flex color camera (3)
  • Zeiss AxioImager motorized fluorescent microscope system with AxioCam camera and dedicated computer and software (4)
  • Zeiss Axiovert 40 CFL fluorescent inverted microscope with AxioCam camera and dedicated computer and software (5)
  • Olympus BH2 light and fluorescence microscope with an attached digital camera and OsteoMetrics, Inc. histomorphometry hardware and OsteoMeasureXP software (v3.2.1.8, purchased in 2011) (6)

Biochemistry, Cellular, and Molecular Biology Sub-Core

The Biochemistry, Cellular, and Molecular Biology Sub-Core provides resources and support for the in vitro analysis of DNA, RNA, or protein isolated from tissues or cells as well as for the performance of various biochemical assays.

Major equipment includes:

  • BioTek Synergy Mx Multi-mode Microplate Reader (BioTek Instruments, Inc.) (1)
    The system features include:
    • Detection methods: UV-Visible Absorbance, Fluorescence, and Luminescence
    • For use with 1- to 384-well plates and PCR plates or cuvette (UV-visible absorbance)
    • Temperature control: 4°C above ambient to 65°C (precision +/- 0.5°C at 37°C)
    • Reading speed: 11 seconds (96 wells), 22 seconds (384 wells)
    • An automated dual-reagent dispenser used for adding solutions at precise times in dynamic and sensitive assays (2)
  • BioTeck Synergy MX Multi-mode Microplate ReaderBio-Rad ChemiDoc XRS+ Gel/Blot imaging system and Image LabTM software (3)
    The system features include:
    • Detection methods: fluorescence, colorimetry, densitometry, chemiluminescence, and chemifluorescence
    • Excitation source: Epi-white light and trans-UV (302 nm)
    • Detection system: Supercooled 16-bit CCD Dynamic range > 4 decades
    • Can accommodate sample sizes up to 28 X 36 cm
  • Two QIAGEN Rotor-Gene Q real-time PCR cyclers (4), one Corbett Rotor-Gene 6000 real-time PCR cycler (5), and one CFX ConnectTM real-time PCR detection system (6)

Surgery RoomsSmall Animal Surgery Sub-Core

Small Animal Surgery Sub-Core provides facilities and support for the performance of small animal surgeries and tissue harvest. Major facilities/equipment include:

  • (1) Two small animal surgery rooms equipped with stainless steel benches, an assortment of light sources, and dissecting microscopes (2)
  • A device for performing closed femoral fractures in mice (Einhorn model) (3)
